Villa was forced to withdraw after 41 minutes as Diego Simeone's side put four past Sociedad to go top of La Liga.
The club has confirmed that an initial scan had shown a "muscle injury on the back of the left thigh".
Villa will undergo further medical tests next week to find out the full extent of the problem.
