Anne Keothavong will meet Germany's Angelique Kerber in round one, while Heather Watson takes on recent Cincinnati Open winner Li Na.
Laura Robson has been pitted against a qualifier in the opening round but could play three-time champion Kim Clijsters in round two.
The US Open will be Clijsters's last Grand Slam before she retires at the end of the year.
