South African Grace carded a 12-under-par round of 60 at Kingsbarns that included 10 birdies and an eagle.
Frenchman Victor Debuisson hit a round of 10-under at St Andrews with Denmark's Thorbjorn Olesen a shot further back.
Sweden's Noren Alexander is one of three men on eight-under also playing St Andrews.
Meanwhile, European Ryder Cup hero Martin Kaymer hit three birdies on his way to a score of two-under at Carnoustie, with England's Oliver Wilson top scoring on the course with five-under.
