The 23-year-old is understood to be one of Mauricio Pochettino's top targets this summer, with recent speculation claiming that the Champions League runners-up are willing to break their transfer record for the second time this window by making a £70m bid.
Spanish outlet AS reports that Lo Celso is keen to move to North London and has asked the club to sanction the deal should a firm offer arrive from Spurs.
Betis are now eager to speed up the process in order to avoid any complications later in the transfer window.
The La Liga outfit are on the verge of signing Borja Iglesias from Espanyol and have been heavily linked with Lyon's Nabil Fekir as a direct replacement for Lo Celso.
Spurs, meanwhile, have already broken their transfer record once this summer with the £63m purchase of Tanguy Ndombele, while Jack Clarke has also joined the club from Leeds United.
