Fly-half Nick Evans's first conversion of the game put Harlequins in front, just one minute before Seb Stegmann's set up his second.
Substitute Scott Hamilton reduced the deficit after 20 minutes, allowing fly-half Jeremy Staunton to convert and then add a penalty to take the lead.
However, Nick Evans's three points meant that Harlequins held on to their lead at the break.
The Quins fly-half added his second penalty of the game after 10 minutes of second-half action.
Staunton responded almost instantly to put the home side just two points behind, but a Joe Marler try after 70 minutes put the game all but out of reach for the Tigers.
